| 【摘要】 目的：研究三七透明质酸钠凝胶对家兔硬膜外瘢痕中环氧化酶-2（cyclooxygenase-2，Cox-2）、转化生长因子β1（transforming growth factor beta-1，TGF-β1）及结缔组织生长因子（connective-tissue growth factor，CTGF）表达的影响。方法：将72只大耳白品系家兔随机分为A、B、C、D 4组，制作家兔L6椎板切除模型，分别在硬膜囊周围滴加生理盐水、三七浓缩液、透明质酸钠、三七透明质酸钠凝胶各0.5ml。各组于术后1、2、4周取手术局部硬膜外瘢痕组织，采用原位杂交法检测Cox-2 mRNA和CTGF mRNA的表达情况，采用免疫组化法检测TGF-β1抗体的表达情况。结果：术后1、2周时D组硬膜外瘢痕中Cox-2 mRNA的表达较其他各组显著减少（P＜0.05），其他各组间无显著性差异（P＞0.05）；各组2、4周时较1周时及A、B、C组4周时较2周时均显著减少（P＜0.05），D组2周和4周时无显著性差异（P＞0.05）。术后1周时CTGF mRNA的表达B、C、D组较A组及D组较B、C组均显著降低（P＜0.05）；2周时D组较其他各组均降低（P＜0.05），其他各组间无显著性差异（P＞0.05）；A、C组2、4周及B组4周较1周时均显著减少（P＜0.05），其他各组各时间点之间无显著性差异（P＞0.05）。术后1周时TGF-β1抗体的表达B、C、D组较A组及D组较B、C组均显著减少（P＜0.05），2周时D组较A、B、C组均显著减少（P＜0.05），其他各组之间无明显差异（P＞0.05）；各组2周与1周时比较均无显著性差异（P＞0.05），4周时较1、2周时均明显减少（P＜0.05）。术后4周时各指标各组之间无显著性差异（P＞0.05）。结论：三七透明质酸钠凝胶可明显抑制硬膜外瘢痕组织中与成纤维细胞增殖相关的Cox-2、TGF-β1、CTGF的表达，其作用优于单独使用三七浓缩液或透明质酸钠。 |

| 【Abstract】 Objective：To investigate the effects of Sanqi sodium hyaluronate gel on the expression of cyclooxygenase-2（Cox-2），transforming growth factor beta-1（TGF-β1） and connective-tissue growth factor（CTGF） in rabbits′epidural scar tissue.Method：72 white rabbits were randomly divided into four groups.After L6 laminectomy，the local dural sacs were soaked by 0.5ml saline（group A），0.5ml Sanqi concentration（group B），0.5ml sodium hyaluronate（group C） and 0.5ml Sanqi sodium hyaluronate gel（group D） respectively.At the 1st，2nd，and 4th week after operation，the expression of Cox-2 mRNA and CTGF mRNA was detected by hybridization in situ and the expression of TGF-β1 antibody was tested by immunohistochemistry in scar tissues.Result：The expression of Cox-2 mRNA in group D was lower than all other groups at the 1st and 2nd week（P＜0.05），and no difference among the other groups at the 2nd week and all groups at the 4th week was noted（P＞0.05）.All the four groups had significant lower expression at the 2nd and 4th week than the 1st week and 4th week in group D（P＞0.05）.The expression of CTGF mRNA in group B，C and D was remarkably lower than group A（P＜0.05） at the 1st week and group D was lower than group B and C as well（P＜0.05）.At the 2nd week group D had lower expression than all other groups（P＜0.05），but no difference among the other groups was noted（P＞0.05）.Group A and C at the 2nd and 4th week as well as group B at the 4th week had lower expression than 1st week significantly（P＜0.05）.No difference among all other groups at all other time points was noted（P＞0.05）.The expression of TGF-β1 antibody in group B，C，D was lower than that in group A remarkably（P＜0.05） at the 1st and that in group D was lower than all other groups at the 2nd week（P＜0.05）.There was no difference among all groups at the forth week（P＞0.05）.There was no difference of each group at the 1st and 2nd week（P＞0.05）.But each group had lower expression at the 4th week than the 1st and 2nd week（P＜0.05）.There was no difference among each group′ expression at the 4th week（P＞0.05）.Conclusion：Sanqi sodium hyaluronate gel can inhibit the expression of Cox-2，TGF-β1，CTGF in rabbits′epidural scar tissue，which is superior to Sanqi concentration or sodium hyaluronate used alone. |
